How to Order HCR™ Gold RNA-FISH

Our webstore is designed to enable you to obtain quotes and place orders at your convenience with an easy-to-use interface and plenty of customizability. The webstore is organized into three components: an HCR™ HiFi Probe, an HCR™ Gold Amplifier, and HCR™ Buffers. Each reagent section includes all the information you need to configure your kit. Additionally, if you already have some reagents on hand, you can easily omit any reagent and order only what you need.

Ordering an HCR™ HiFi Probe
  1. Select Sample Type
    • Choose the sample type that best matches your experiment. HCR™ Buffers are optimized on a sample type basis, so it is critical this selection is accurate to enable high performance.
  1. Select Organism
    • Choose the organism you are working with from the drop-down menu. The organisms listed here are part of our Infinite Catalog that contains full transcriptomic information already. If it's not listed, type it in to create a custom entry as shown below: 
  1. Enter Target Information
    • Start by typing in the name of your target. If your organism is part of the Infinite Catalog and the target is part of the latest NCBI annotated database, the target information should auto-complete.
    • Otherwise, please enter a custom target name and sequence.
  1. Select the HCR™ System for Your HCR™ HiFi Probe
    • Choose the HCR™ System you would like to use with your HCR™ HiFi Probe. This step ensures that the probe you receive is compatible with the corresponding HCR™ Gold Amplifier. For instance, HCR™ HiFi Probes designed for the X1 system are only compatible with X1 HCR™ Gold Amplifiers (i.e., X1-488, X1-546, and X1-647).
      • Note: While most HCR™ Gold Amplifiers are available in single fluorescent channels, the X1, X2, and X3 systems offer additional flexibility by supporting labels in 488, 546, and 647, providing enhanced versatility for target screening.
  1. Specify the Target # of Binding Sites
    • The default option is Standard, which requests up to 20 binding sites along the target RNA sequence and is recommended for most targets and sample types. For applications requiring enhanced signal sensitivity, the Boosted option is available, aiming to design up to 40 binding sites. This setting is ideal for targets or samples where a signal boost is needed, such as single-molecule imaging of low-abundance targets.
  2. Select the Scale
    • Choose the scale of the HCR™ HiFi Probe you wish to order. The indicated number of experiments is based on the selected sample type.
Ordering an HCR™ Gold Amplifier
  1. If you've already chosen your preferred system for the HCR™ HiFi Probe in the previous section, the HCR™ Gold Amplifier will automatically populate based on your selection. If not, choose your desired HCR™ Gold Amplifier system and label from the options available in the drop-down menu.
    • Note: If you selected an X1, X2, or X3 system and wish to use it with a different fluorophore (e.g., pairing X1 with 546 or 647), you can adjust your selection using the drop-down menu under Label.
  1. Choose the scale of the HCR™ Gold Amplifier you wish to order. The indicated number of experiments is based on the selected sample type.
Ordering HCR™ Buffers
  1. The final step in completing your HCR™ Gold RNA-FISH order is to select the desired volumes of HCR™ Buffers you'd like to order.
  2. For additional details on how many experiments you can perform based on the volume of buffers you order, click the question mark icons next to each buffer type.

If you have any special instructions regarding probe design, order fulfillment, or other key details, please include them in the Comments section. For example, if you have a specific transcriptome you'd us to use for probe design, let us know how to access it.

Once all reagents are added to your cart, you can easily review and make any final edits directly on the cart page. When you're ready, proceed to checkout to place your order and receive a quote by email. Note that no charges will be made at this stage. Once we receive a valid PO or credit card payment, we'll begin probe design and production - kits typically ship within 10-15 business days.
